My two cents: ultimately, at the heart of teaching is a human connection. By definition, teaching involves transferring knowledge from one mind to another, stimulating thought, encouraging critical thinking. The best online courses are merely vehicles for facilitating this process.
With this in mind, I'd answer that the biggest problem/challenge is successfully teaching/stimulating/inspiring in an increasingly remote and disconnected world. Digital fatigue and low-attention viewing are real (and constantly growing) problems. Online course creators have to look for ways to not only overcome technical challenges but to make a real connection. This requires expertise, of course, but also originality, technical innovation and that genuine human connection.
